You could do this with Dovecot deliver and a Sieve script.

I'm not sure what define a message readed (a mark, a xheader or something) thats why I'm asking here!

If you're not using deliver, then it depends on if you're using maildir or mbox. With maildir the flags are in the filenames. With mbox you can do it by adding (or replacing) "Status: R" header.
