See: http://wiki2.dovecot.org/AuthDatabase/PasswdFile the syntax is: user:password:uid:gid:(gecos):home:(shell):extra_fields so your user file is wrong. But this error would come later.
The config shows that you have not configured no passdb. Re-check that you've included the appropriate auth*ext files, e.g. the auth-passwdfile.conf.ext.
